Determination of Estradiol-17β concentration using a competitive ELISA assay

Determination of Estradiol-17β concentration, an important female sex hormone mainly produced in the ovaries and involved in reproduction, bone growth, brain development, and regulation of the menstrual cycle, by using a competitive ELISA assay. The fundamentals of ELISA techniques, hormone quantification, and the importance of estradiol measurement in reproductive health and biomedical research were evaluated. Explanation of the principle of a competitive ELISA, where estradiol present in the sample competes with enzyme-linked estradiol for binding to specific antibodies. In this assay, a higher estradiol concentration in the sample results in a lower color intensity and lower absorbance reading, creating an inverse relationship between hormone concentration and absorbance. An experiment using microplate wells, standards with known estradiol concentrations, antibodies, washing buffers, and a chromogenic substrate was performed by the students. After incubation and washing steps, absorbance is measured at 405 nm using a spectrophotometer. In the second part of the class, students analyze the data by correcting absorbance values, calculating averages, determining the percentage of maximum binding (B/B0), building a standard curve, and estimating the estradiol concentration of an unknown sample.
